Principles
Bio-Layer Interferometry (BLI) is a label-free biosensing technology based on optical interference principles. Biomolecules are immobilized on sensor tips and dipped into analyte-containing solutions; molecular binding causes changes in the interference spectrum at the sensor tip, enabling real-time monitoring of binding kinetics.
Features
- No labeling required — does not affect native molecular activity
- Real-time monitoring of entire binding/dissociation process
- Simple operation with relatively high throughput (multi-channel parallel detection)
- Good sample buffer compatibility
Typical Applications
- Antibody affinity maturation and screening
- Protein-protein interaction kinetic analysis
- Small molecule-target binding characterization
- Concentration quantification analysis
