Principles
Olink Multiplex Proteomics is built on Proximity Extension Assay (PEA) technology. A pair of specific antibodies recognizes and binds the target protein, each conjugated with a DNA oligonucleotide. When both antibodies bind the same protein, the DNA strands hybridize and extend, forming a DNA barcode quantifiable by qPCR or NGS.
Core Advantages
- Ultra-high sensitivity: Detection limit down to fg/mL level, suitable for complex biofluids like plasma/serum
- High throughput: Single panel covers 5 to 5,400+ protein biomarkers
- Wide dynamic range: Linear detection range spanning several orders of magnitude
- Minimal sample: Only 1–6 µL of biofluid required
Typical Applications
- Inflammatory disease subtyping and mechanism research (IBD, pancreatitis, etc.)
- Tumor immunotherapy patient stratification and efficacy prediction
- Neuropsychiatric biomarker discovery (depression, early dementia screening)
- Drug safety biomarker monitoring
