ASO Antisense Oligonucleotide Synthesis

2026-09-13

Overview

Antisense Oligonucleotides (ASOs) are short nucleic acid molecules that regulate gene expression through Watson-Crick base pairing with target mRNAs. We provide end-to-end services from sequence design to scale-up synthesis.

Chemical Modification Options

  • Backbone modification: Phosphorothioate (PS), Boranophosphate (BP)
  • Sugar modification: 2'-OMe, 2'-F, 2'-MOE, LNA (Locked Nucleic Acid)
  • Terminal modification: 5'/3' conjugation (cholesterol, GalNAc, PEG, etc.)
  • Mixed backbone: Gapmer design (DNA gap + modified wings)

Synthesis Specifications

  • Length range: 12–30 nt
  • Scale: 10 µg – 10 g
  • Purity grade: HPLC purified (> 95%) or PAGE grade
  • QC standard: LC-MS MW confirmation + HPLC purity analysis
